Abstract

Modified, discriminative method of staining of mucosal mast cells and granulated intraepithelial lymphocytes has been setup and tested. The method included: a) fixation either in Carloy’s fixative followed by post-fixation with 10% formalin, or in methanol: formalin: glacial acetic acid; b) staining with toluidine blue, pH 1, and c) counterstaining with mordant-acid fuchsin. Qualitative assessment of granules of mucosal mast cells and granulated intraepithelial lymphocytes as well as of their nuclei, is highly discriminative after fixation and staining with this method, Well contrasted pink stained nuclei with mordant-acid fuchsin facilitates the recording of positions of the cells in any quantitative analysis.
